What the B Vitamins Do

Thiamin, riboflavin, niacin, pantothenic acid, and biotin belong to the water-soluble B-vitamin family alongside vitamin B6, vitamin B12, and folate. As a group, B vitamins act as coenzymes in the enzymatic processes that support nearly every aspect of cellular function, though each one plays its own distinct role.[2]

Thiamin (B1) works in the body as thiamine pyrophosphate (TPP), a cofactor for enzymes including pyruvate dehydrogenase, alpha-ketoglutarate dehydrogenase, transketolase, and branched-chain ketoacid dehydrogenase.[1] TPP sits at crucial steps in the citric acid cycle and pentose phosphate pathway, playing a major role in how the body metabolizes glucose for energy.[2]

Riboflavin (B2) is a cofactor in redox reactions through two active forms, FAD and FMN, that shuttle electrons during cellular energy production.[1] Those active forms are also needed to synthesize niacin, folate, vitamin B6, and heme proteins, and riboflavin itself is involved in metabolizing carbohydrate, protein, and fat.[2] Niacin (B3) functions through NAD+ and NADP+, a related pair of redox coenzymes, and the body can synthesize some niacin from dietary tryptophan.[1]

Pantothenic acid (B5) is a building block of coenzyme A and fatty acid synthase, and it's essential for synthesizing cholesterol, fatty acids, and acetylcholine, the neurotransmitter involved in muscle activation and memory.[1][2] Biotin (B7) is necessary for the metabolism of protein, fat, and carbohydrates, catalyzing reactions across fatty acid, glucose, and amino acid metabolism.[1][2]

The five vitamins' coenzyme roles are documented across two independent lines of research: a StatPearls biochemistry review covering the water-soluble vitamins as a class, and a 2022 review in The Permanente Journal focused on B-vitamin functions in medicine.[1][2] Both describe niacin's and riboflavin's active forms as central to redox chemistry, the electron-transfer reactions cells depend on for energy production.[1][2]

Riboflavin's role turns out to be interconnected with the other B vitamins: its active forms are needed to synthesize niacin, folate, vitamin B6, and heme proteins, so its chemistry supports work the other B vitamins ultimately depend on too.[2]
